
The Cleared Tissue DISPIM (CT-DISPIM) is a version of the DISPIM that is specifically optimized to image large samples of cleared or expanded tissue. Like the DISPIM, it is a multi-view light sheet microscope that acquires two orthogonal image volumes and computationally fuses them into a single, high-resolution image volume. As a light sheet microscope, imaging is very fast, with cm scale samples being acquired within a few hours.
Two pairs of objectives are available for imaging: 17X 0.4NA (isotropic resolution ~1um) and 25X 0.7NA (isotropic resolution ~0.5um). The 0.4NA objectives can accommodate samples as large as 5mm x 2cm x 3cm while the 0.7NA objectives can accommodate samples as large as 2mm x 2cm x 3cm.
Cleared tissue sample preparation and the resulting terabyte scale data processing can be extraordinarily challenging, so AIM staff are available to help you with both. We have extensive knowledge of common clearing protocols (iDISCO, CUBIC, PEGASOS) and routinely process and analyze terabyte light sheet data on Biowulf.
